Nicotinamide Riboside Prevents Low Kidney Nephron Numbers Caused by Maternal Protein Restriction: Preclinical Findings
Synopsis
Babies born with fewer nephrons (kidney filters), often due to poor maternal nutrition, have higher risk for kidney disease and high blood pressure later. There are no current treatments to increase nephron numbers. This study showed that in mice born to mothers on a low-protein diet, kidney SIRT3 (a protective enzyme) levels were low, and nephron number was reduced. Giving nicotinamide riboside (NR), a NAD+ precursor that activates SIRT3, restored SIRT3 levels and activity, improved kidney structure, and protected mitochondria from damage. These findings suggest that boosting SIRT3 with NR during kidney development could prevent nephron loss and its long-term health effects.
