Nicotinamide Riboside Reduces Inflammation and Supports Mitochondrial Health: Preclinical Findings

Activation of the NLRP3 inflammasome, a key driver of inflammation linked to metabolic dysfunction, can be influenced by nutrient levels and mitochondrial health. In a study of healthy volunteers, fasting for 24 hours reduced NLRP3 activity compared with the fed state. Laboratory experiments showed that the sirtuin enzyme SIRT3 protects mitochondria and limits inflammasome activation, while loss of SIRT3 increases mitochondrial stress and inflammation. Activating SIRT3, including with nicotinamide riboside (NR), improved mitochondrial function and decreased NLRP3 activity in immune cells. These results suggest that nutrient control and NR-mediated SIRT3 activation may help regulate inflammation and support metabolic health in humans.
