Nicotinamide Riboside Improves Memory and Learning in Alzheimer's Disease: Preclinical Findings


Synopsis

NAD+ is an important coenzyme that supports energy production in mitochondria and is linked to longevity and protection against Alzheimer’s disease (AD). Its precursor, nicotinamide riboside (NR), can raise NAD+ levels and activate PGC-1α, a protein that helps break down BACE1, the enzyme that produces toxic beta-amyloid (Aβ) plaques. In this study, researchers gave NR to Tg2576 mice, a model of AD, for three months. They found that NR increased brain NAD+ levels, improved memory and learning, and strengthened synapses. NR also boosted PGC-1α, which promoted BACE1 breakdown and reduced Aβ buildup. Cell experiments confirmed that these benefits depended on PGC-1α activity. Overall, the study suggests that NR may help protect the brain from Alzheimer’s-related cognitive decline by reducing Aβ production and supporting healthy brain metabolism.
