Synopsis

NAD+ is a central redox cofactor, but its role in activating sirtuins—a family of NAD-dependent proteins that optimize mitochondrial function—has made it a focus of longevity research. Because sirtuin activity depends on NAD+ availability, increasing NAD+ levels through precursors or by blocking its breakdown (via PARPs or CD38/CD157) can activate these proteins and promote healthy aging. This review examines how NAD+ metabolism interacts with sirtuin function, how NAD+ levels shift in different physiological states, and how manipulating these pathways could serve as a therapeutic strategy.
