Safety Evaluation of Nicotinamide Riboside: Preclinical Findings


Synopsis

Nicotinamide riboside (NR) is a natural form of vitamin B3 found in small amounts in foods. A synthetic version, Niagen™, was tested for safety using a series of lab and animal studies. These included genetic toxicity tests and studies on rats over 14 and 90 days. NR showed no genetic toxicity and no deaths even at very high doses (5000 mg/kg). The 90-day study compared NR to nicotinamide, a related vitamin B3 form. Both had similar safety profiles, with some effects seen in the liver, kidneys, ovaries, and testes at the highest doses. The no-adverse-effect dose was 300 mg/kg/day, while some mild effects appeared at 1000 mg/kg/day. These results support NR's safety for use in humans at appropriate doses.
