Nicotinamide Riboside Protects the Intestinal Barrier From Heat Stress: Preclinical Findings


Synopsis

Heat stress damages the intestinal epithelial barrier, allowing harmful substances to enter the body. This study tested whether nicotinamide riboside (NR), a NAD+ precursor, could protect the gut during heat exposure. Mice given NR for 10 days showed increased intestinal NAD+ and NADH, and when exposed to heat, NR prevented the drop in NAD+/NADH ratio, reduced barrier dysfunction, and lowered inflammatory and oxidative markers including IL-6, TNF-α, and TBARS. Heat stress also reduced mtDNA copy number and ATP levels, but NR did not improve these mitochondrial deficits. Overall, NR protects the gut barrier during heat exposure primarily by reducing inflammation and oxidative stress, but not by restoring mitochondrial function.
