Nicotinamide Riboside Improves Mobility in Mitochondrial DNA Disease: Clinical Findings

Objectives

To evaluate safety, physical function, and mitochondrial responses to NR supplementation.

Journal

Genome Medicine

Key Outcomes

  • NR supplementation significantly, but minimally, improved the timed "up and go" test from 11.9 to 9.96 seconds.
  • NR tended to improve measures of mitochondrial function in muscle, most prominently in those with large-scale mitochondrial DNA deletions. Several muscle measurements moved in a direction consistent with improved mitochondrial function and a lower proportion of deleted mitochondrial DNA.

Duration

4 weeks

Dose

1250 to 2000 mg

Study Design

Open-label, single-arm exploratory study in 8 adults with genetically confirmed heteroplasmic mitochondrial DNA (mtDNA) disease.

Diagnoses were either with mutations at m.3243A>G/T or single large-scale mtDNA deletion.
