Nicotinamide Riboside Improves Mitophagy and Reduces Neuroinflammation in Ataxia Telangiectasia: Preclinical Findings
Synopsis
In the premature aging disease ataxia telangiectasia (A-T), cells show mitochondrial dysfunction and a senescence state that causes inflammation (SASP). This is linked to buildup of damaged mitochondria and DNA in the cell cytoplasm, activating inflammatory pathways via STING. Boosting NAD+ with nicotinamide riboside (NR) helps clear damaged mitochondria through mitophagy, which depends on the protein PINK1. NR treatment reduced brain inflammation, prevented nerve cell loss, and improved movement in A-T mice. This suggests that fixing mitochondrial problems by increasing mitophagy with NR could be a therapy for A-T and related aging conditions.
