Synopsis

Non-rapid eye movement sleep (NREMS) is linked to lower brain glucose use, which slows metabolic reactions involving NAD+. This study tested whether boosting NAD+ with nicotinamide riboside (NR) affects sleep regulation in mice. Long-term dietary NR reduced time spent in NREMS by 17% and sped up the recovery of sleep need. These results suggest that increasing NAD+ availability enhances the brain's energy capacity, reduces sleep pressure, and affects cortical activity. Overall, the study shows that NAD+ and redox metabolism play a key role in sleep homeostasis and brain function.
