Nicotinamide Riboside and Nicotinamide Mononucleotide Restore Nerve Function in Diabetic Neuropathy: Preclinical Findings


Synopsis

Reduced NAD+ signaling contributes to nerve degeneration in diabetic peripheral neuropathy. Researchers tested NAD+ precursors, nicotinamide riboside and nicotinamide mononucleotide, in cultured neurons and diabetic mice. Both precursors increased NAD+ and SIRT1 activity, promoting neurite growth in neurons. In diabetic mice, NR and NMN improved sensory function, restored nerve conduction, and preserved intraepidermal nerve fibers. The benefits were dependent on SIRT1, highlighting NAD+ supplementation or SIRT1 activation as promising strategies to treat DPN.
