Nicotinamide Ribosides Improves Aerobic Performance: Preclinical Findings
Synopsis
Nicotinamide riboside (NR) is a form of vitamin B3 that helps increase NAD+ and improve mitochondrial function, but its effect on physical fitness is unclear. In a study with sedentary and trained mice, NR was given daily for 5 or 10 weeks. Exercise training involved treadmill running five days a week. While NR alone did not improve aerobic performance, it did raise NAD+ levels in muscle. When combined with exercise, NR enhanced aerobic capacity more than exercise alone. This improvement was linked to increased amounts of enzymes involved in NAD+ production and mitochondrial proteins. Genetic analysis supported that the enzyme NMNAT3 correlates with mitochondrial health and exercise traits. Additionally, NR plus exercise increased the number of slow-twitch (type I) muscle fibers, which are better suited for endurance. Overall, these results indicate that NR, especially when combined with aerobic training, can boost muscle metabolism and improve endurance.
