Effects of Nicotinamide Riboside on Energy Metabolism and Exercise Performance: Preclinical Findings

Nicotinamide riboside (NR) is a form of vitamin B3 that can boost NAD(P) levels, which are important for energy metabolism in cells. While previous studies in older or sick mice showed that NR improves health and muscle function, researchers tested it in healthy young rats and found that NR slightly reduced exercise performance. In this study, rats were given NR for 21 days, and some underwent an exercise test. NR increased certain liver energy molecules (NADPH and glycogen), but also caused signs of oxidative stress in the blood and lowered activity of protective antioxidant enzymes. It also reduced blood glucose and lactate levels. These results suggest that in healthy individuals, taking NR may not always be beneficial and could potentially have mild negative effects on metabolism and exercise performance.
