Low NAD+ levels have been linked to many health problems related to aging and lifestyle. Supplements with NAD+ precursors (vitamin B3 forms) may help, and nicotinamide riboside (NR) has shown strong results in animal studies for improving health and treating disease. This review looks at how NAD+ works, what causes NAD+ shortages, NR's path from discovery to clinical testing, current limits of NR-based therapies, and ways to improve them, with the goal of choosing the most effective NAD-boosting approach.
