Nicotinamide Riboside Prevents Hearing Loss in Cockayne Syndrome: Preclinical Findings


Synopsis

Age-related hearing loss (ARHL) is one of the most common disorders affecting elderly individuals. There is an urgent need for effective preventive measures for ARHL because none are currently available. Cockayne syndrome (CS), a premature aging disease, causes early hearing loss similar to age-related hearing loss. Using mouse models of CS, researchers found that NAD+ levels drop in the cochlea. Short-term treatment with nicotinamide riboside (NR), an NAD+ precursor, prevented hearing loss, restored outer hair cells, and improved cochlear health. NR also reversed deficits in synaptic ribbons crucial for hearing, likely by supporting the NAD(H)-dependent protein Ribeye. These results point to NR as a potential treatment for hearing loss in CS and aging.
