Safety Evaluation of Synthetic Nicotinamide Riboside (NR-E) in a 90-Day Oral Toxicity Study: Preclinical Findings


Synopsis

Nicotinamide riboside (NR) is a form of vitamin B3 that boosts NAD+ levels and may help counter age-related declines in cellular energy. This study tested the safety of a high-purity, synthetic NR (NR-E) in rats over 90 days at doses up to 1,200 mg/kg/day. No deaths or significant health issues were observed, though male rats at the highest dose had a small reduction in body weight. Overall, NR-E was well-tolerated, and the no-observed-adverse-effect-level (NOAEL) was determined to be 500 mg/kg/day for males and 1,200 mg/kg/day for females.
