Nicotinamide Riboside Supplementation Improves Post-Ovulatory Oocyte Quality: Preclinical Findings

As eggs age after ovulation, their quality declines, reducing the chances of successful fertilization and embryo development during assisted reproductive technology (ART) procedures like IVF. This study found that nicotinamide riboside (NR), a vitamin B3–derived NAD+ booster, helps preserve the quality of aging oocytes during in vitro culture. In mouse experiments, NAD+ levels naturally decreased in post-ovulatory eggs, leading to mitochondrial dysfunction, oxidative stress (ROS buildup), and DNA damage. Supplementing with NR restored NAD+ levels, maintained healthy chromosome and spindle structure, reduced ROS and apoptosis, and improved early embryonic development. These results suggest that NR supplementation may enhance egg viability and embryo quality, potentially improving the success rate of fertility treatments in women undergoing ART.
