Nicotinamide Riboside Protects Against Liver Fibrosis: Preclinical Findings

Nicotinamide riboside (NR) boosts NAD+ levels and offers protective effects in various diseases, but its role in liver fibrosis is unclear. This study tested NR’s ability to protect against liver fibrosis in mice and liver cells. Mice with liver fibrosis induced by CCl4 were treated with NR, which prevented and even reversed fibrosis. In liver cells, NR stopped the cells from activating and causing damage by activating Sirt1 and lowering certain protein modifications. When Sirt1 was blocked, NR’s protective effect was reduced. These results show that NR protects against liver fibrosis by suppressing harmful cell activation, highlighting NR’s potential as a therapeutic agent for liver fibrosis.
