Nicotinamide Riboside Prevents SARM1-Induced NAD+ and ATP Depletion: Preclinical Findings


Synopsis

Mitochondrial dysfunction is a key feature of neurodegenerative diseases like Parkinson’s. SARM1 promotes nerve damage by breaking down NAD+, and its activity is increased when phosphorylated at serine 548 by JNK under oxidative stress. This leads to reduced mitochondrial function and ATP levels, making neurons more vulnerable. Notably, supplementation with the NAD+ precursor nicotinamide riboside (NR) suppressed SARM1-induced NAD+ and ATP loss in a dose-dependent manner. These results suggest that abnormal SARM1 activation contributes to neuronal energy failure, and boosting NAD+ with NR may help protect neurons in Parkinson’s and other neurodegenerative diseases.
