Nicotinamide and Nicotinamide Riboside Induce Rapid Differentiation and Function in Retinal Pigment Epithelial Cells: Preclinical Findings

Human retinal pigment epithelial (RPE) cells, especially the ARPE-19 cell line, are commonly used in eye and general epithelial cell research. While they are convenient and consistent, these cells often do not fully develop the specialized features of natural RPE cells. Researchers developed a simple and fast method to culture ARPE-19 cells so they show better cell shape, cytoskeleton organization, and RPE-specific functions. They found that nicotinamide and nicotinamide riboside were crucial for promoting this improved differentiation, highlighting the role of metabolism in RPE cell development.
