Nicotinamide Riboside Extends Lifespan and Improves Health in Ataxia Telangiectasia Models: Preclinical Findings

Ataxia telangiectasia (A-T) is a rare inherited disease causing worsening nerve damage and loss of coordination. It is caused by problems in the ATM gene, which controls how cells repair DNA damage. The exact reasons for nerve and brain cell loss in A-T aren't fully understood. This study found that neurons lacking ATM have increased PARylation, low NAD+ levels, and mitochondrial dysfunction. Boosting NAD+ levels with NR improved symptoms in cell, mouse, and worm models by helping DNA repair and clearing damaged mitochondria. These treatments improved nerve function, slowed memory loss, and extended lifespan. The findings connect DNA damage and mitochondrial problems as key causes of early aging in A-T and suggest NAD+ boosting as a potential therapy.
