Synopsis

Damage from oxidative stress is a major cause of hearing loss, particularly through injury to inner hair cell (IHC) synapses, which connect sensory cells to the auditory nerve. This study found that activating SIRT3, a mitochondrial enzyme that controls reactive oxygen species (ROS), can protect these cells from damage. In lab experiments using hydrogen peroxide to mimic oxidative stress, researchers observed that nicotinamide riboside (NR)—a vitamin B3 derivative and NAD+ booster—increased SIRT3 activity and helped preserve hair cell connections. NR treatment restored mitochondrial energy production, reduced ROS buildup, and increased levels of protective proteins like FOXO3a, SOD2, and IDH2. These effects prevented cell death and improved mitochondrial health. The findings suggest that boosting SIRT3 through NR supplementation may help protect against hearing loss caused by oxidative or age-related damage.
