NAD+ Precursors in Health, Disease, and Aging: Review


Synopsis

NAD+ is an essential molecule that helps the body make energy, send signals between cells, keep mitochondria healthy, and decide whether cells survive or die. Research shows that NAD+ levels drop with age, likely because the body makes less and uses more of it. In animal studies, restoring NAD+ levels has been shown to extend healthy lifespan and improve conditions like premature aging and neurodegenerative diseases. This suggests that boosting NAD+ with compounds called precursors could help prevent or treat age-related diseases such as Alzheimer's. This review looks at the biology, safety, and possible side effects of key NAD+ precursors, including nicotinamide riboside (NR), nicotinamide mononucleotide (NMN), and the recently discovered dihydronicotinamide riboside (NRH). Overall, NAD+ precursors show great promise for supporting healthy aging and fighting metabolic and neurodegenerative diseases.
