Impact of Nicotinamide Riboside and β-Hydroxybutyrate on Lifespan: Preclinical Findings

Researchers studying the longevity of C. elegans (a model organism for aging research) found that nicotinamide riboside (NR)—a vitamin B3–derived NAD+ booster—and beta-hydroxybutyrate (BHB)—a ketone body and histone deacetylase (HDAC) inhibitor—each extend lifespan but through different life stages. NR promoted longer life when given during larval development, while BHB extended lifespan only when given during adulthood. Surprisingly, combining NR and BHB shortened lifespan, suggesting that both compounds activate similar stress-response and longevity pathways that, when overstimulated, become harmful—a phenomenon known as hormesis. The study highlights how timing and dosage are critical when using metabolic compounds like NR and BHB to influence longevity and mitochondrial health.
