Beyond Pellagra—NAD+ Deficiency in Aging and Disease: Review


Synopsis

Interest in NAD+ has surged because low NAD+ levels are linked to many health problems, including metabolic diseases, neurodegeneration, reproductive disorders, cancer, and aging. Researchers are now focusing on moderate but still harmful NAD+ deficiencies, not just severe cases like pellagra. This includes developing accurate models to study NAD+ biology and testing new ways to boost NAD+ inside cells. This review summarizes early landmark research, new experimental systems for studying NAD deficiency, and emerging strategies to restore healthy NAD+ levels.
