Stem Cell Extracellular Vesicles Loaded with Nicotinamide Riboside, NAD+, and Resveratrol Rejuvenate Skin and Prevent Photoaging: Clinical Findings

Objectives

To investigate the potential synergistic effects of loading small extracellular vesicles from adipose (fat)-derived stem cells (ASC-sEVs) with NR, NAD+, and resveratrol on photoaging and skin rejuvenation in immortalized human skin cells and three human subjects.

Journal

Skin Research and Technology

Key Outcomes

In Vitro:

  • In ultraviolet B (UVB) radiation-exposed immortalized human skin cells, the loaded vesicles containing NR, NAD+, and resveratrol reduced oxidative stress more effectively than individual ingredients or vesicles alone.

In Vivo:

  • Topical application of the loaded vesicles was safe, with no signs of irritation or allergic reactions.
  • Topical application of the loaded vesicles containing NR, NAD+, and resveratrol to the back of the left hand improved skin texture, increased hydration by 19%, improved elasticity by 104%, and reduced pore volume by 51%.

Duration

8 weeks

Dose

2 μg*

*Loaded vesicles contained 1 μg sEVs (∼1.21x109 particles), 2 μg NR, 1 μg NAD+, 1 μg resveratrol in 0.1 mL of 0.9% sodium chloride per dose.

Study Design

Prospective, placebo-controlled study in 3 subjects with no history of skin allergy or skin abnormalities
