Nicotinamide Riboside Supplementation Reverses Aging-Associated NAFLD: Preclinical Findings
Synopsis
Aging increases the risk of non-alcoholic fatty liver disease (NAFLD), but the mechanisms are not fully clear. This study tested whether nicotinamide riboside (NR), a precursor to NAD+, could prevent aging-related NAFLD in mice. Aged mice were given NR in their food for three months. NR restored NAD+ levels, reduced fat accumulation, lowered cholesterol and triglycerides, and improved liver enzyme levels. It also reduced liver steatosis, fibrosis, and inflammatory cell infiltration, including Kupffer cells. These results suggest that oral NR supplementation can reverse aging-associated NAFLD-like symptoms and may help prevent liver disease in older individuals.
