Nicotinamide Riboside Slows Biological Aging by 2.5 Years: Clinical Findings

Objectives

To investigate the effects of NR supplementation and high-intensity interval training on skeletal muscle epigenetic aging, as assessed by DNA methylation–based epigenetic clocks and measures of epigenetic age acceleration.

Journal

Aging Cell

Key Outcomes

  • NR supplementation slowed biological aging in three of the seven clocks, including a roughly 2.5-year reduction in age acceleration on the muscle-specific MEAT clock.
  • Exercise produced different, clock-dependent changes, including higher DunedinPACE estimates.

Duration

  • NR: 5 months
  • Exercise: 4 or 6 weeks

Dose

  • NR: 1000 mg
  • Exercise: High-intensity interval training, three times per week

Study Design

Exploratory analysis of three independent cohorts: a nonrandomized, open-label, parallel-assignment study of identical twin pairs with BMI discordance (one leaner, one heavier), with 30 participants providing paired muscle samples, and two exercise cohorts of 39 and 37 participants.
