Nicotinamide Riboside Reverses Metabolic Dysfunction in Werner's Syndrome: Preclinical Findings


Synopsis

Werner syndrome (WS) features premature aging and metabolic dysfunction, yet mechanisms remain unclear. This study found that loss of the WRN protein accelerates early fat cell formation in both stem cells and zebrafish, partly through upregulation of SMARCA5, which regulates gene activity. Restoring WRN normalized SMARCA5 levels and fat cell development. Importantly, treatment with the NAD+ precursor nicotinamide riboside (NR) improved fat metabolism in both models, suggesting NR could help correct metabolic dysfunction in WS and potentially other aging-related conditions.
