Nicotinamide Riboside Protects Optic Nerve from Degeneration: Preclinical Findings


Synopsis

Nicotinamide riboside (NR), a precursor to NAD+, protects optic nerve axons from damage caused by tumor necrosis factor (TNF). In rats, NR reduced axon loss, improved autophagy markers (increased LC3-II, decreased p62), and increased SIRT1 levels in the optic nerve. The enzyme NRK1, which helps convert NR to NAD+, was found in retinal ganglion cells and optic nerve fibers, and NR treatment boosted its expression. Blocking SIRT1 partially reversed NR’s protective effects, suggesting that NR protects axons by enhancing NAD+ metabolism, SIRT1 activity, and autophagy.
