Nicotinamide Riboside Protects Against DNA Damage in Cells By Increasing NAD+: Preclinical Findings


Synopsis

Mitochondrial cytochrome c oxidase (COX, complex IV) helps cells make energy. When COX is deficient, it can cause neuromuscular problems and sometimes anemia. Researchers found that COX4-1 and COX6B1 deficient cells had much higher levels of DNA damage. Experiments with cyanide showed DNA damage can happen even when energy and ROS levels are normal, suggesting it’s not caused by stress or energy loss. Treatment with nicotinamide riboside (NR) reduced DNA damage, while blocking NAD+ with a PARP inhibitor made it worse. This suggests that NAD+ depletion, not energy loss, may cause DNA damage in COX deficiency, and NR could help protect cells.
