Nicotinamide Riboside Protects Against Diet-Induced Liver Injury: Preclinical Findings

Nicotinamide riboside (NR), a precursor to NAD+, helps prevent metabolic and age-related disorders in mice, but the role of naturally occurring NR metabolism is less understood. Researchers studied mice lacking NR kinase 1 (NRK1), an enzyme essential for NR to boost NAD+ in the liver. Mice without NRK1 showed reduced glucose production, impaired mitochondrial function, and worse metabolic health. When fed a high-fat diet, these mice developed glucose intolerance, insulin resistance, fatty liver, and DNA damage in the liver. This shows that endogenous NR metabolism via NRK1 is critical to maintain liver NAD+ levels and protect against diet-induced metabolic problems, suggesting NRK1 could be a target for treating metabolic disorders.
