Nicotinamide Riboside Dose Improves Metabolic Flexibility: Preclinical Findings


Synopsis

Metabolic flexibility—the ability to switch between burning carbs and fats—is a marker of metabolic health. To study NR's effect on this, mice on a mildly obese diet received varying NR doses for 15 weeks. The best metabolic flexibility, measured by changes in respiratory exchange ratio (RER) when switching fuels, occurred at 30 mg/kg NR. This dose also increased expression of adipogenesis regulator Pparγ and antioxidant genes (Sod2, Prdx3) in white fat tissue. Higher or lower NR doses had smaller or no benefits. Thus, 30 mg/kg NR appears optimal for supporting metabolic health in this obesity model.
