Nicotinamide Riboside and Pterostilbene Safely Elevate NAD+ Levels in Humans: Clinical Findings

Objectives

To evaluate the safety, tolerability, and efficacy of nicotinamide riboside and pterostilbene (NRPT) supplementation on sustaining elevated whole-blood NAD+ levels in healthy older adults.

Journal

npj Aging and Mechanisms of Disease

Key Outcomes

  • NRPT supplementation was safe, well-tolerated, and significantly increased whole blood NAD+ levels.
  • NRPT was associated with a significant reduction in alanine aminotransferase (ALT), a marker of liver damage, although it also led to a small increase in total and LDL cholesterol.
  • Functional performance improved with NRPT, as evidenced by gains in the 30-second chair stand and the 6-minute walk test, suggesting that prolonged supplementation may support overall muscle health.

Duration

12 weeks

Dose

250 or 500 mg NR + 50 or 100 mg PT

Study Design

Randomized, double-blind, placebo-controlled, multi-site study in 120 healthy, older adults
