Natural Nicotinamide Riboside–Loaded Stem Cell Extracellular Vesicles Synergistically Combat Aging: Preclinical Findings

Synopsis

This study explored the potential of small extracellular vesicles (ASC-sEVs) from adipose-derived stem cells for skin care and anti-aging. Researchers purified ASC-sEVs and analyzed their bioactive components, including miRNAs, proteins, and lipids, to understand how they regulate skin health. They then combined ASC-sEVs with various natural compounds like nicotinamide riboside (NR), resveratrol (RES), vitamin C (VITC), retinol (RET), and arbutin (ARB) to test their effects on skin cells exposed to UVB radiation. The results showed that the compound-loaded ASC-sEVs were more effective than the compounds or ASC-sEVs alone in reducing oxidative stress, preventing wrinkles, and decreasing melanin production. In particular, combinations of NR, RES, and VITC reduced reactive oxygen species (ROS) levels, while RES and RET helped reduce wrinkle-related proteins. Additionally, RES and ARB lowered melanin content in skin cells. Importantly, all combinations were non-toxic to the cells. This study highlights the promising potential of ASC-sEVs combined with natural compounds for enhanced anti-aging skin treatments.
