NAD+ Precursors Prevent Allergic Responses by Regulating Immune Function: Preclinical Findings

Nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN)—both vitamin B3 derivatives that boost NAD+ levels—may help prevent severe allergic reactions such as anaphylaxis. In this study, researchers found that increasing NAD+ in mast cells (immune cells responsible for allergic responses) reduced their activation and release of inflammatory molecules. In mice, NR or NMN treatment significantly decreased symptoms of anaphylaxis, including swelling and inflammation. The protective effects relied on SIRT6, a protein that depends on NAD+ to regulate cell activity. These findings suggest that NAD+ precursors like NR and NMN could be developed into therapies to control severe allergic and inflammatory reactions by keeping mast cell activity in check.
