The Biology and Therapeutic Potential of Nicotinamide Riboside and Nicotinamide Mononucleotide: Review

Synopsis

Research on NAD+ biology has surged, especially around two intermediates—nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN)—which show promise in preventing and treating age-related conditions. While their exact metabolism and tissue-specific behavior are still being clarified, studies suggest that their effectiveness depends on where NAD-related enzymes, nucleotidases, and transporters are most active. A new framework now links NAD+ metabolism directly to aging and longevity, and the challenge ahead is to translate strong preclinical results into proven health benefits for humans.
