NAD+ Precursors Nicotinamide Riboside and Nicotinamide Regulate Cardiac Sodium Channels: Preclinical Findings


Synopsis

The heart’s electrical activity depends on sodium channels (NaV1.5) and dysfunction can cause dangerous arrhythmias. Researchers studied how NAD+ precursors nicotinamide riboside (NR) and nicotinamide (NAM) affect NaV1.5 currents in cells and heart tissue. NR increased peak sodium current and reduced harmful late sodium current, improving channel function through multiple mechanisms including Sirtuin 1-dependent deacetylation. NAM had less of an effect. Mice fed NR showed improved cardiac electrical activity. These findings suggest NR could be a new treatment for heart arrhythmias caused by sodium channel defects.
