Effects of Nicotinamide Riboside on Hearing: Preclinical Findings


Synopsis

This study examined how repeated high-dose intravenous nicotinamide riboside (NR)—a vitamin B3–derived NAD+ booster—affects hearing in Wistar rats. Researchers measured the activity of both the cochlea (inner ear) and auditory nerve pathways using auditory evoked potentials and otoacoustic emissions before and after NR treatment. After administering a cumulative dose of 2700 mg/kg, the results showed that repeated NR exposure negatively affected the cochlear structures, which are responsible for converting sound vibrations into nerve signals. These findings suggest that while NR is generally considered safe at nutritional levels, excessive or long-term high-dose use may harm auditory function, highlighting the importance of dosage regulation in future research and clinical applications.
