Safety of Intravenous Nicotinamide Riboside Across Multiple Doses in Rats: Preclinical Findings


Synopsis

This study evaluated the safety of repeated intravenous nicotinamide riboside (NR) at varying doses in Wistar rats. NR was well-tolerated at 150, 300, and 450 mg/kg, with no adverse effects on the heart or autonomic ganglia. However, the 600 mg/kg dose caused significant toxicity, including cardiac, liver, and kidney damage, reduced cardiovascular performance, lower exercise tolerance, and increased mortality. These findings indicate that high-dose IV NR can be harmful, while lower doses appear biocompatible and safe.
