Nicotinamide Riboside Rejuvenates Aged Hematopoietic Stem Cells and Improves Regeneration: Preclinical Findings

With age, hematopoietic stem cells (HSCs) lose their ability to regenerate and remain quiescent, leading to an expanded stem cell pool that can cause blood disorders. Researchers found that nicotinamide riboside (NR), a form of vitamin B3, restores youthful metabolic function in HSCs by improving mitochondrial health, reducing metabolic gene expression, and decreasing mitochondrial mass and network size. Continuous NR treatment shifts the aged HSC transcriptome closer to that of young cells, improves bone marrow composition, and enhances regenerative capacity in transplants. Thus, NR supplementation may promote healthy aging by rejuvenating the hematopoietic system.
