Nicotinamide Riboside and Metabolic Cofactors Improve Gut Health in Fatty Liver Disease: Preclinical Findings


Synopsis

The gut barrier controls what passes into the body and protects organs like the liver. An unhealthy diet and lifestyle cause gut microbiome imbalances (dysbiosis) and gut barrier damage, which worsen non-alcoholic fatty liver disease (NAFLD). This study tested a combination of metabolic cofactors (betaine, N-acetylcysteine, L-carnitine, nicotinamide riboside) in mice with diet-induced NAFLD. Treatment improved gut structure, reduced gut leakiness, lowered inflammation, and increased gut cell growth. It also changed gut bacteria composition and beneficial short-chain fatty acids. These effects helped slow NAFLD progression by improving gut-liver communication.
