Synopsis

Parkinson's disease (PD) involves mitochondrial dysfunction, reduced proteasome activity, and dopamine depletion. NR activated mitochondrial function in C. elegans, rescuing α-synuclein-related defects. In mouse models with proteasome inhibition, NR initially improved behavior but later reduced dopamine and related gene expression in the substantia nigra. These results suggest NR can improve mitochondrial health, but long-term effects on dopamine need careful evaluation.
